45' Willys MB

1945 MB, Have to put gas directly into carburetor to start. Once it starts and runs for a while it will start without putting gas directly into the carburetor. If it sits over night it won't start without putting gas directly into carburetor again. It seem like it looses its prime. I've tried using the priming pump on the gas pump. Any suggestions?

To me it sounds like a leaky fuel bowl. are there any stains/ oily deposits around the carb?

Let it sit for a couple of days then go directly to the upper carb housing and remove it and see just how much gas is still in the bowl. None = bowl leak. Also you didn't mention what effect if any cranking with full choke had.

Cranking with full choke didn't help.

After sitting for two days there was still plenty of gas in the bowl. Nothing leaking. Still would not start without putting gas directly into carburetor.

Sounds like clogged passageways. Do you have the stock Carter WO carb installed? If you do then start your troubleshooting with the proper Army TM 9-1826A, Chapter 6 page 50 where you can review the carb's system operations and then go to Chapter 4 Troubleshooting on page 25 and get started.
